Nein, glaube ich nicht. Höchstens He is not ambitious of assuming office, aber das wäre doch auch komisch. Was ganz anderes wäre bsp. He is not ambitious enough to assume this office.

You can't be "ambitious to do something"

But you can be ambitious enough to do sth., and that's what #2 suggests, although I also agree with REScan that it does sound a bit funny in this particular context.

Yes, but the meaning is different.

"He has no ambitions to" = he doesn't plan to do sth.

"He is not ambitious enough to" = he's not hungry enough for honor / power to do sth.
